Abstract: A tripartite chimeric recombinant spidroins iN7RC was designed based on two spidroin genes from Araneus ventricosus, successfully expressed in E. coli cells, and obtained a highly pure 162. 5 ku recombinant spidroin through purification at a yield of 34 mg/ L.The circular dichroism (CD) spectra of iN7RC spidroin solution under different pHs indicated that the content of the secondary structure α-helix was distinctly decreased along with the declining pH value. Recombinant spider silk fibers with superior strength and extensibility were acquired by hand-drawing, which paved the foundation for the extensive application of biomaterials.
